(Transfection
in the presence of 10% fetal bovine serum using NovaFECTOR)

|
1. Grow cells in 35mm
dish using a medium
that contains 10% fetal bovine serum to
55-70% confluence.
2. Aspirate old medium from dish.
|
|
|
|
Add plasmid DNA and
NovaFECTOR lipid to 1.0 ml of serum free medium (in 1.5 ml
siliconized microtube). Let sit at room temperature for 15
minutes to allow DNA:lip complex formation. (Start with 2-3
ug of DNA and 8-10 ug of lipid, then change the concentration
of DNA and/or lipid to optimize)

3. Pipet 1.0 ml of medium
contains
20% fetal bovine serum to dish.

|
|
|
|
|
|
|
|
|
|
|
|
|
|
|
|
4. Add DNA:lipid complex
to dish.

|
|
|
|
|
|
|
|
|
|
|
5. Incubate to express
reporter gene product.
